Prospection of innovative therapeutic alternatives for infectious diseases with relevance in Dentistry: low temperature atmospheric pressure plasma and nanoencapsulate of bioactive substance

Abstract

Some infectious diseases represent a great challenge in Dentistry due to their increasing prevalence, low number of therapeutic options and/or increase in the antimicrobial resistance. Considering that the searching for new therapies is highly needed, this project aims to prospect innovative therapeutic alternatives, evaluating the applicability of low temperature atmospheric plasma (LTAPP) and nanoencapsulates of the bioactive substance ellagic acid (NAE) in the treatment of oral candidosis and periodontal disease. Pursuing this objective, the project will be divided into 3 subprojects: 1) Potential of LTAPP and NAE in the treatment of oral candidosis: the ideal physical conditions for planktonic and biofilms of C. albicans will be determined. The ellagic acid in the concentration with antifungal activity (determined in previous study) will be complexed in cyclodextrin and chemically characterized by differential scanning calorimetry, scanning electron microscopy and analysis of dissolution profile. After in vitro evaluation, the effectiveness of the therapies will be investigated in vivo in induced oral candidosis murine model. For elucidation of possible mechanisms of action, the effects on the integrity of C. albicans cell wall and plasmatic membrane, ergosterol level, morphogenesis, production of exoenzymes, adherence to epithelial cells and expression of SAPs, HWP1 e ALS will be investigated. 2) Potential of LTAPP in the treatment of periodontal disease (PD): The effect of LTAPP on the main bacterial species involved in PD will be evaluated by qPCR. The effects on the biofilm-host interaction and tissue response will be analyzed by proteomic analysis (in vitro), by bone resorption and by cytokines involved in the process (in vivo). 3) Analysis of the cyto and genotoxic potential of LTAPP and NAE in the effective conditions for the treatment of oral candidosis and periodontitis: The analysis will be performed by assays of cell viability, TUNEL, metanuclear alterations, micronuclei and DNA ploidia, effect on cell cycle, detection of ROS and genotoxicity in microspheres model. The toxicity of NAE will be also evaluated using Drosophila melanogaster model. Data will be analyzed statistically by evaluating the normality of data distribution followed by the suitable specific analysis. Alpha level will be set at 0.05. (AU)
