Evaluation of antimicrobial potential in vitro and in vivo of Piper umbellatum extracts incorporated or not into nanostructured lipid carriers

Abstract

Piper umbellatum L (Piperaceae) is a species of great pharmacological interest because have been shown that presents, among many activities, great antimicrobial potential, which has already been investigated by our group (FAPESP Process 2013 / 25243-3; 2013 / 10578-0; 2013 / 23680-7). The incorporation of plant extracts in nanostructured lipid systems can enhance their biological activity.To advance our research the objective of this study is to evaluate the antimicrobial activity of P.umbellatum extracts incorporated and not, into lipid nanostructured systems. The project is linked to the development of the doctoral degree of Vanessa Raquel Greatti under my supervision. Nanostructured lipid systems will be developed in the Laboratory of Pharmacotechnique/FCF and evaluated to the encapsulation efficiency and controled liberation monitored. The antimicrobial activity will be evaluated using the bacterial and yeasts strains and the dermatophytes Trichophyton rubrum and Microsporum canis. MIC (Minimum Inhibitory Concentration), MBC and MFC (minimum bactericidal concentration and minimum fungicidal concentration) of the extracts and extracts incorporated in lipid nanostructured systems will be evaluated by microdilution in microplates The cytotoxicity assay will be performed using murine macrophages and human keratinocytes for evaluation of toxicity. Experimental dermatophytoses models in vivo will be evaluated for antifungal activity of the extracts increased or not in lipid nanostructured systems. (AU)
