Abstract

Klebsiella pneumoniae is a Gram-negative opportunistic pathogen. This bacterium has become a worldwide public health problem in recent years, mainly because of the emergence of hypervirulent and multidrug-resistant clinical isolates capable of producing invasive infections not only in immunocompromised patients but also in healthy individuals. Bacteria control gene expression in a coordinated manner, whether in response to stressful conditions or during pathogen-host cell interactions at the time of infection. In bacteria, an important step on gene expression regulation occurs during transcription, a process that involves Sigma factors and regulatory proteins. In several pathogenic bacteria, the Sigma factor E promotes the transcription of stress-responsive genes and virulence genes either involved in resistance to phagocytosis or related to modulation of the inflammatory response of macrophages. However, little is known about the role of K. pneumoniae Sigma factor E on the interaction with host cells, and the relevance of this Sigma factor in animal models of infection. Thus, this study aims to investigate the role of Klebsiella pneumoniae Sigma factor E in the processes of adhesion, invasion, phagocytosis and inflammatory responses in host eukaryotic cells and in the induction of lung infection in an animal model. The results from this study may contribute to a better understanding of the role of Sigma factor E at the Klebsiella pneumoniae pathogenicity. (AU)
