Determining the role of cementocytes in dental cementum homeostasis

Abstract

Cementocytes not only share morphological features with osteocytes, but also express several common biomakers. However, in contrast to osteocytes, there is a lack of evidence on the potential biological properties of cementocytes. Thus, we hypothesized that cementocytes play an important role in dental cementum homeostasis. In order to test our hypothesis, a comparative and systematic gene expression analysis will be performed using LCM/RNA-Seq on cementocytes in areas of active matrix apposition. In addition, morphological and ultrastructural changes in cementocytes will be investigated by transmission electron microscopy and confocal microscopy in samples labeled with fluorescein isothiocyanate. Lastly, based on our previous experience obtaining a cementocyte-like cell line from mouse (IDG-CM6), we aim at obtaining and characterizing cementocytes from human teeth in order to develop an in vitro model to propose functional assays. Based on our findings it is expected to define the role of cementocytes in dental cementum homeostasis and also identify potential biomolecules to be used in order to control dental cementum formation and regeneration.
