Structural and functional analysis of the fibronectin type III domain (FnIII) of one beta-glucosidase belonging to the family GH3: interaction with large polymeric substrates and thermostability


The enzymatic hydrolysis of cellulose to glucose requires mainly three types of cellulases working synergistically: endoglucanases, cellobiohydrolases and Beta-glucosidases. The endoglucanases and cellobiohydrolases are often inhibited by cellobiose. Thus, Betaglucosidases are important in terms of avoiding decreased hydrolysis rates of cellulose over time due to cellobiose accumulation. A pretreatment process is frequently employed prior to the enzymatic hydrolysis reaction to remove lignin. Previous studies have showed that the addition of lignin to pure cellulose can downgrade sugar release by a factor of 60%. Thermotoga petrophila is a hyperthermophilic bacterium that produces a Beta-glucosidase belonging to the family GH3 (TpBGL3). TpBGL3 is an enzyme composed of three distinct domains. The N-terminal domain is an (Alpha/Beta)8 triose phosphate isomerase barrel, followed by a five-stranded Alpha/Beta sandwich domain. The C-terminal domain has a fibronectin type III fold whose function remains unknown, although it may be involved in the process of anchoring the enzyme on large polymeric substrates and in thermostability. In this project, we propose to conduct functional and structural studies of the domain FnIII from TpBGL3. The main objective of this project is to characterize the possible interaction of the FnIII domain with large polymeric substrates as lignin and cellulose. Besides, we will study the role of the FnIII domain on the thermostability and also dimerization. The information obtained in this project may be useful for biotechnological applications in the biofuel industries. (AU)
