ImagestreamX Mark II (Amnis) proposal for the acquisition of the ImagestreamX Mark II equipment which combines flow cytometry techniques with optical fluorescence microscopy. Whole blood samples or isolated cells are labeled with immunofluorescent molecules and the ImageStreamX equipment, used in combination with the IDEAS software, will allow the determination of the mean fluorescence intensity of each cell simultaneously with an image of the same cell in the bright-field and the relevant channel fluorescence channels. This equipment provides the opportunity to link fluorescence data with data regarding the morphology of the cell, providing the most consistent, reliable data. The ImageStream cytometer combines the speed capabilities, sensitivity and phenotyping of flow cytometry with detailed image microscopy and functional insights. This unique combination allows for a wide range of applications that would be impossible with isolated techniques. Applications include equipment for research; intracellular signaling, cell-cell interactions, cell morphology, cell cycle, autophagy, cell death and cell differentiation, among others. Currently, to our knowledge, there is no other center that has this equipment in the state of São Paulo. We believe the acquisition of this equipment would be of great benefit to the UNICAMP and the Hemocenter (Blood Center) at UNICAMP, as well as to FAPESP, as the Hemocenter has a flow cytometry sector with a team of highly qualified employees. Additionally two researchers from our group (Kleber Fertrin and Venina Dominical) performed internships at the NIH, USA using the same equipment (Fertrin et al, Blood 2014; 124:... 658-60; Dominical et al, Haematologica 2014 2014 Nov; 99 (11): e214-7). We are sure that all of the Research groups, at the Hemocenter, UNICAMP, will use this equipment extensively and we will make the equipment available to users at UNICAMP and other universities using the Hemocenter's website.
