Abstract

Brevipalpus-transmitted viruses (BTV) are known since early XXth century, but detailed knowledge about them only became available in the last 15 years. Great efforts were made especially on the most economically important among BTV, the Citrus leprosis virus C (CiLV-C) which resulted in the understanding of its molecular nature, relationship with the host plants and the vector and its epidemiology. Consequently, a rapid and precise method for virus detection and more efficient and economical management of these viruses were developed. One important finding about BTV was that there are at least two distinct types of BTV: the cytoplasmic (BTV-C) and nuclear type (BTV-N) and that these viruses cause consistently only localized infection, without becoming systemic in the infected plant. It became also clear the existence of numerous plant species, naturally infected by BTV. However, these advances brought up new challenges, as to understand the level of diversity of these BTV-C and BTV-N, and also to understand the taxonomy and diversity of Brevipalpus populations, the mechanisms of virus transmission, management of the mite by biological control, etc. This project intends to solve some of these questions, through an integrated action, with the participation of a group of researchers of different specialities, from several institutions, expecting to give an additional and significative advance on the knowledge of this important and peculiar group of plant viruses. This photomicroscope, to be purchased, will permit examination of samples under direct illumination, differential (DIC) and phase contrast, rotating stage and UV illumination for fluorescence and immuno-fluorescence Works. An eficiente and friendly digitalized image capture system with high resolution is attached to the microscope. In the Project it will be used for:a. Studies on Brevipalpus transmitted viroses (BTV) / vector relationship. especially feeding process (subproject 3) b. Evaluation of the morphological variations in diferente populations of Brevipalpus in slide mounted samples (subproject 4). c. Analysis of anatomical changes in leaf lesions caused by BTV, and the use of immunofluorescence techniques to detect viral antigens and observe their distribution in the lesions tissues (subproject 6 and 7).Although the chosen instrumento (Axio Imager 2, produced by Carl Zeiss) has the highest price among the 3 presented budgets, we justify this selection because: i. NAP/MEPA has a former model of Zeiss photomicroscope (Axioskop 2), aquired through a Fapesp grant (1998/16311-3) in 1998, thus 15 years old. The grant holder was prof. Sergio F.Pascholatti (Dept.Fitopatol. & Nematol.) who kindly permitted that this instrument was installed in the NAP/MEPA for a multiuser system, and indeed it was intensively used by users not only from ESALQ, but also from several other research institutions. It is still workable, but with obvious signs of wearing. ii. Zeiss has one of the best maintainance system for microscopes in Brazil. iii. The chosen Zeiss microscope has one of the best optical system and image capture system and friendly for use. iv. In the UV illumination system for fluorescence Zeiss developed UV lamps of new generation which can last up to 2.000 h v. Finally, some parts of the old Axioskop is interchangeable with the new instrument. This microscope will be part of NAP/MEPA facilities and will be available for all the scientific community of Esalq and other research institutions, and the nucleus will be in charge of its maintainance.
