Abstract

Many researches on periodontal disease have sought a correlation between local and systemic factors. In Aggressive Periodontitis (AP), an exacerbated inflammatory response to some microorganisms has been reported, however, still little is known about the mechanisms linking these factors. The objectives of this study will be to evaluate some cytokines in gingival crevicular fluid (GCF) and serum of patients with AP (localized and generalized) and healthy subjects. In addition to the clinical data, samples will be obtained from peripheral blood and GCF of two disease sites for analysis of the cytokines and from subgingival biofilm to detect and quantify, through the Real-Time PCR, four periodontal pathogens: Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, Tannerella forsythia, and Treponema denticola. Furthermore it will be do a research for A. actinomycetemcomitans clone JP2 due to its strong association with AP. Kits for detection of human cytokines (Human Luminex Screening Assay - R&D Systems Inc., Minneapolis, USA)) will be used for detection of cytokines in GCF and serum in order to verify the existence of an association between local and sera mediators in healthy subjects and patients with AP. The analyzed cytokines will be: IL-1², IL-8, IL-10, IL-11, IL-17, ICAM-1, MCP-1, M-CSF and GM-CSF, among others. The hypothesis is that there is a correlation between cytokines related to AP found in GCF and serum and that the serum could serve as predictors of disease. (AU)
