Effect of cooling system in bovine semen cryopreservation

Abstract

One of the main steps in the sperm cryopreservation process is the cooling step. The rate of cooling can reduce the thermal shock and damages sperm cells with that in cryopreservation better results could be improved by using curves were slower and homogeneous cooling, but in some refrigeration systems, such as common refrigerators, it does not. Thus, the objective is comparing different systems cooling and cryopreservation from bovine semen and its effects on the quality of cryopreserved semen. The experimental design is randomized in a 5x2 factorial arrangement with 5 cooling systems and 2 cryopreservation, totaling 10 treatments. Two ejaculates of 15 Nelore bulls totaling 30 ejaculates collected by electroejaculation will be used. The ejaculates be diluted in a single fraction (BotuBov ®) to a final concentration of 60x106 spz/ml after dilution are cooled for 5 hours at 5 different cooling systems (TK 4000 ® with cooling curve of 0.25°C/min, 4000 TK ® with cooling curve of 0.5°C/min, refrigerator Minitüb® 518C , BotuTainer® and other common refrigerators) and 2 cryopreservation (TK 4000 ® and box with liquid nitrogen) . Sperm kinetics will be performed by the device IVOS (Version 14 , Hamilton - Thorne Bioscience ) at three times: after 5 hours of cooling (before cryopreservation), cryopreservation and after the test quickly thermotolerance (TTR) . Moreover, the evaluation of the integrity of the plasma, mitochondrial membrane and acrosomal association using fluorescent probes is performed. The analyzes of the data generated will help elucidate which better system cooling and cryopreservation that may benefit the technicians working in the field. (AU)
