Abstract

Recent results from our group indicated that mutants of Salmonella enterica for genes encoding NAPs (nucleoid-associated proteins) are virulence attenuated and able to induce, in different extension, protection in the mouse model of infection. These results suggest an important role of NAPs in bacterial virulence. Based on this, the aim of this project is further study the role of different NAPs in the biology of S. enterica Typhimurium and Enteritidis. New mutants will be constructed and phenotypically evaluated with regard growth in vitro, acid-resistance, bile salts, invasion, persistence and multiplication in macrophages and biofilm formation. Virulence attenuation and immunogenicity will be evaluated in the murine model of infection. Also, high-throughput methodologies will be employed to study gene expression. The results will bring new information on the role of NAPs in S. enterica virulence. In addition, attenuated mutants of S. enterica will be evaluated regarding the therapeutic effect on tumors in the mouse model. (AU)
