Abstract

An enormous challenge in medical stem cell research is to direct the differentiation toward specific fates in vitro. This comes from presenting cells with the correct balance of biochemical and mechanical cues on engineered substrates that mimic the native matrix. In vivo, the extracellular matrix is highly dynamic and subject to constant remodelling that regulates and affects stem cell differentiation fate. For this purpose, Dr. Ram Sharma will join our research group to collaborate in the study of biomedical systems based on biodegradable/biocompatible PHBHV nanoparticles that mimic the extracellular matrix. Then, PHBHV based nanoparticles with different sizes, will be synthesized through nanoprecipitation and/or emulsion solvent evaporation, followed by their surface functionalization with amino groups (-NH2). The functionalized nanoparticles will be used in another project coordinated by Prof°. Dr. Ram Sharma, in which such polymeric systems should perform as substrates in the cellular differentiation of mesenchymal stem cells. This study will allow understanding how the properties of nanoscale substrates can interfere in the cellular differentiation mechanism. (AU)
