DESI-HPTLC-Bioautography: New strategies to rapid Identification of anti-mastotos bioactive metabolites from actinomycetes.

Abstract

Microorganisms, in particular the actinomycetes are known to produce a vast number of bioactive secondary metabolites of interesting pharmaceuticals and agrochemical industry. Rapid detection of bioactive compounds plays strategic' role in their natural products investigation. TLC is one of the most frequently used chromatographic methods for separating complex chemical compounds. Several ESI-related ambient ionization methods have been used to efficiently and rapidly characterize chemical compounds separated on TLC plates. Hyphenated techniques involving mass spectrometry combined with thin-layer chromatography (TLC-MS) and high-performance column liquid chromatography (HPCLC-MS) provide on-Jine extensive structural information on the metabolites prior to isolation. Mass spectrometry (MS) has become a powerful and wide range technique in analytical and bioanalytical analysis. This overwhelming success and broadness has resulted mainly from the unmatched abilities of MS to detect, count, and characterize atoms and molecules of many types, compositions, and sizes. Two recent advancements in ionization mass spectrometry, namely electrospray ionization (ESI) and matrix-assisted laser desorption ionization (MALDI), have been largely responsible for the rapid growth of this field over the last fifteen years. Since its debut in 2004 there has been explosive growth in the applications and variants of ambient MS, and the proliferation of almost thirty new techniques in that time, often combining separate desorption and ionization processes in a single method.
