In vivo evaluation of lipoteichoic acid, endotoxin and microrganisms in necrotic teeth

Abstract

The present study will evaluate the levels of lipoteichoic acid (LTA ), endotoxins and microorganisms in root canals with pulp necrosis and radiographically visible periapical lesion during treatment with endodontic instrumentation mechanized and 2.5% sodium hypochlorite. In addition, there will be a correlation between the amount of LTA, endotoxins, microbial organisms, periapical lesion size and symptomatology. 20 uni-root teeth with necrotic pulp will be instrumented with the system of alternating rotation Reciproc and irrigated with 2.5% sodium hypochlorite. Samples will be collected of root canal immediately after opening coronary after biomechanical preparation, after using trisodium EDTA and 17% after 14 days of action from the dressing of calcium hydroxide with 2% chlorhexidine gel. For all collections will be performed the following tests: identification and quantification of microorganisms by Real Time PCR, quantification of endotoxin testing by kinetic chromogenic lysate from amoebocytes of Limulus and quantification of lipoteichoic acid by enzyme immunoassay (ELISA). The periapical lesions will be measured in early endodontic treatment, for later correlation with the amount of endotoxins, lipoteichoic acid and symptomatology. The data will be statistically analyzed using the Kruskall-Wallis, Dunn (5% significance level) and Linear Correlation of Pearson. (AU)
