The Toll Like Receptors gene polymorphisms(TLR) involved in the inflammatory process of Helicobacter pylori and gastric carcinogenesis and assessment of changes in gene expression


Gastric cancer represents a global health problem, with a neoplasm in which genetic and environmental factors are involved. H. pylori is a bacterium that colonizes the stomach and affects about 50% of the world population. The gastric infection by this microorganism has been strongly linked to the etiology of gastric carcinogenesis, since the active host inflammatory response, which may lead to chronic inflammation. Thus, it has been evaluated the association of genetic polymorphisms of factors involved in the inflammatory process which can modulate the pattern of immune response of the host. This is the case known as Toll like receptors (TLR) that recognize molecular patterns associated with the microorganisms, including TLR2 (bacterial lipoprotein receptor) and TLR9 (bacterial primers receiver) are involved in the response to infection by H. pylori. Given the scarcity of studies of polymorphisms of these genes in dyspeptic patients and its possible involvement in the pathogenesis of H. pylori and gastric carcinogenesis, we proposed to evaluate by PCR-RFLP TLR2 polymorphisms (rs3804099 and deletion -196 to -174) and TLR9 (rs5743836, rs187084) and in dyspeptic patients with gastric cancer and a group of individuals whose histopathology was considered Normal. In order to draw a profile of these genetic polymorphisms associated with increased risk for gastric lesions found and infection by H. pylori, as well as assess the expression levels in the gastric mucosa by PCR in real time and the possible association of these SNPs in the level of protein expression of the receptors. The molecular epidemiology studies and susceptibility to diseases are being intensified, so have been relevant to the understanding of individual variability and predisposition to cancer. (AU)
