In vitro evaluation of the antimicrobial action of denture cleansers solutions

Abstract

The objective of this study is to evaluate, through in vitro analyses, the effectiveness of cleansing chemical methods - alkaline hypochlorites, alkaline peroxides and castor bean solution (Ricinus communis) - regarding the antimicrobial action against specific microorganisms. From quadrangular metallic matrices (10mm x 2mm), it will be made 800 specimens of heat-polymerized acrylic resin, which will be contaminated with micro-organisms Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans, Bacillus subtilis, Escherichia coli, Streptococcus mutans, Enterococcus faecalis and Candida glabrata and immersed (20 minutes) in the following solutions: 1) Positive Control - Saline, 2) Experimental I: sodium hypochlorite 0.25%, 3) Experimental II: sodium hypochlorite 0.5%, 4) Experimental III: sodium hypochlorite 1%, 5) Experimental IV: castor bean solution 10%, 6) Experimental V: alkaline peroxide solution Kirawa; 7) Experimental VI: alkaline peroxide solution Efferdent; 8) Experimental VII: alkaline peroxide solution Kukident. After this period, the specimens will be rinsed in saline and placed in test tubes containing 5.0 ml of Letheen medium. After stirring, 50mL of the resulting suspension of the serial dilutions (10, 10-¹ 10-² and 10-3) will be seeded in Petri dishes with selective culture media for each microorganism and incubated at 37°C for 24h. After incubation, the number of colonies of each duplicate will be counted and the values of cfu/ml calculated. The obtained data will be processed according to the log 10 formula and statistically analyzed. As additional analysis it will be also examined the cell viability by fluorescence microscopy. (AU)
