Putative role of nitric oxide and greline as mediators/modulators of the febrile response in broiler chicks


Birds and mammals are able to develop febrile response to pyrogenic agents, such as lipopolysaccharide (LPS) from Gram negative bacteria. While there are several studies about mechanisms involved in mammalian fever, including the approach of the role of nitric oxide (NO) and ghrelin in such response, the avian fever is much less studied. Recent data from our laboratory demonstrated that the inhibition of NO production in the central nervous system (CNS) decreases body temperature (Tb) of 5-day old broiler chicks, indicating an opposite effect of that observed in mammals, in which NO acts on the CNS to induce heat loss responses in different stress conditions, such as fever, physical activity, heat stress and anapyrexia. Regarding the hormone ghrelin, it is known that it stimulates food intake and inhibits fever in mammals. In contrast, ghrelin inhibits food intake in ducks, chicks and quails, but no data exists about its role in febrile response in birds. Thus, in the present study it will be investigated the role of NO and ghrelin in LPS-induced fever in 5-day old broiler chicks. To this end, it is proposed the following protocols: i) dose-response curve of the effect of intramuscular (im) injection of LPS in Tb; ii) effect of im injection of L-NAME (NO production inhibitor) in LPS-induced fever; iii) measurements of nitrite-nitrate concentrations (end products of NO metabolism) in plasma and diencephalon of febrile chicks; iv) effect of intracerebroventricular (icv) injection of L-NAME in LPS-induced fever (oxygen consumption and Tb); v) dose-response curve of the effect of icv injection of ghrelin in Tb; vi) effect of icv injection of ghrelin in Tb and food intake after LPS injection; viii) effect of icv injection of ghrelin in oxygen consumption after LPS injection.
