Periodontal disease progression monitoring through the analysis of molecular markers

Abstract

The aim of the study is to monitor the periodontal effects and characterize the progression of periodontal disease, in active sites with progressive attachment loss, through the analysis of gene expression and proteins present in differents types of samples (saliva, gingival fluid and gingival biopsies) from the same patient/site. Thirty patients will be grouped in one of two groups: control group (C; n = 10) with healthy patients and test group (DP; n = 20) those with diagnosis of periodontitis. The patients will be submitted to the complete periodontal examination, the collection of the samples and to the conventional periodontal treatment. After two months the patients will be reappraised and new samples collection will be carried out. The clinical parameters (biofilm, bleeding on probe, probing depth and clinical attachment) will be used for diagnosis of the clinical status and, based on tolerance method, for the identification of the active sites with progressive attachment loss. The samples will be analyzed through the polymerase chain reaction in real time (Real-Team PCR) and the immunoenzimatic assay (ELISA) to establish the gene expression and proteins of interest related to the inflammation, immune-regulating, destruction of collagen, bone loss and healling. The data will be grouped and submitted to the statistical analysis for verification of differences between the groups C and DP and between the inactive and active sites, and still establish correlation with the molecular profile and between the different samples.
