Nitric oxide releasers based on ruthenium carboxylates and their interaction with some relevant biomolecules

Abstract

This project proposes the synthesis and characterization of trinuclear ruthenium compounds (clusters) of general formula [Ru3O(CH3COO)6(L)2(NO)]PF6 (L = solvent molecules or pyridinic ligands) aiming to develop nitric oxide releasing species, in a controlled fashion, by means of light irradiation or by reductive reactions, in order to amplify the initial knowledgement reported in literature, which is limited to two papers focusing these compounds for biological applications. In this context, analogues with VO instead of NO may be studied in order to compare to the photoinduced liberation of NO and also the carbonyl compounds may present interesting properties from the biological view - point. This proposal will cover an important gap in the actual literature, which is the synthesis of a new and larger series of ruthenium nitrosyls based on the trinuclear clusters in such a way that, in the next two years, the group will have the opportunity to study, understand and report the behavior of the compound as a novel class of NO releasers. Besides that, we intend to expand the focus of the research performed in our group, by means of the investigation of the complexes reactivity towards biomolecules such as aminoacids, endogenous reductants, human serum albumin and ct-DNA.
