Next generation phylogeography of the Pilosocereus aurisetus cactus species group: hypotheses testing and markers development

Abstract

Despite the high diversity and levels of endemism in the xeric enclaves scattered throughout the Brazilian Cerrado biome, the mechanisms of diversification in these environments are still poorly understood. Among the plant taxa ecologically restricted to these xeric enclaves, the group of cactus species PILOSOCEREUS AURISETUS is composed by eight species exhibiting disjunct distribution in southeastern and central-western Brazil. An intricate phylogeographic structure has been observed in this group, which was probably the result of a complex history of demographic events related to changes in species ranges and secondary contact with gene flow. However, the investigation of these events was largely hindered by the insufficient variation in the molecular markers available, which retain very weak signals of historical demographic processes occurring in these species. This project aims to generate genomic data using Roche 454 pyrosequencing from a sample of 40 individuals across eight populations from four species utilizing a procedure of reduced representation library and individual MID barcodes. The sequence data (complete loci and SNPs) will be used in coalescent and population structure analyses to test particular phylogeografic hypotheses concerning the diversification process of such species. The genomic data will also be used to the development of anonymous nuclear (ANMs) and microsatellite (SSRs) markers appropriate to be used in subsequent phylogeographic studies encompassing extensive population sampling in this species group.
