Effects of the use of probiotics in the treatment of periodontal disease in animals experimentally induced immunosupressed with chemotherapy DRUG(5-FLU): a histometric and immunohistochemical study in rats

Abstract

The present study aims to evaluate histomorphometrically and immunohistochemically the effects of medication use probiotic adjunct procedure to scaling and root planing (SRP) in the treatment of experimentally induced periodontal disease in immunosuppressed animals with chemotherapy drug (5-fluorouracil). One hundred twenty-six animals are subjected to induction of periodontal disease by adaptation of a cotton thread around the first molars. Immunosuppression is achieved by intraperitoneal injection of 5-fluorouracil (Fluorouracil, 50 mg / ml) which will be implemented in two phases: during installation ligation (- 7 days) and 48 hours (- 5 days) after installation ligation. Seven days after onset and progression of periodontal disease induced, the bandage is removed and the animals were randomly divided into six groups of 21 animals each to receive the following treatments: Group I (n = 21) - animals systemically healthy, will not suffer any Local or systemic treatment, Group II (n = 21) - animals systemically healthy, local treatment will not suffer, only systemic treatment with saline, Group III (n = 21) - animals treated systemically with 5-fluorouracil (5-FLU) not suffer any local treatment and Group IV (n = 21) - animals treated systemically with 5-FLU, not suffer treatment of scaling and root planing, receive local treatment with probiotic (PRO), Group V (n = 21) animals treated systemically with 5-FLU, receive treatment with scaling and root planing (SRP) and Group VI (n = 21) animals treated systemically with 5-FLU treated with SRP and local treatment with Probiotic (RAR-PRO). The first molar to contralateral disease that is induced experimental animals of group III, will suffer no local treatment and will be used to assess the systemic effects of the chemotherapy drug without periodontal disease induced. The animals, seven in number for each group are sacrificed at intervals of 7, 15 and 30 days. Parts of the experimental side (with induction of disease) and the contra-lateral area of the specimens of Group III (no periodontal disease) are processed for histomorphometry and immunohistochemistry.
