Analysis of the lipodemic profile and bioavailability of natural and bio-transformed flavonoids in rats

Abstract

Earlier reports on our research group have shown that hydrolysis of the glycoside fraction of rutin enhance its antioxidant and anti-proliferative activities as measured by in vitro tests. Enzymatic hydrolysis with total or partial liberation of the glycosidic fraction is capable of generating simpler compounds with enhanced functionality. This study is designed to make comparative assessments of the effects of two flavonoids (rutin and hesperidin) on the plasma lipids profile and the biochemical parameters involved in the oxidative processes before and after enzymatic modification. For test purposes, the compounds (1mmol eq flavonoid/Kg animal live weight) will be administered orally to male Wistar rats for 30 days. Plasma lipids, erythrocytes and tissues will be extracted using chloroform and methanol, the classes of phospholipids separated by solid phase extraction (SPE) chromatography and the fatty acids composition determined by gas chromatography (GC). Plasma concentration of aglycone will be analyzed using ultra performance liquid chromatography coupled with mass spectrometry (UPLC-MS). Additionally, biochemical-molecular procedures will be undertaken to quantify malondialdehyde, antioxidant enzymes and expressions of desaturases (FADS1 and FADS2) in the animals' plasma and tissues. This proposal is intended to contribute to obtaining greater understanding of the effects of flavonoids and their hydrolyzed derivatives and to elucidate those effects in the light of the compounds' structures.
