Cytotoxicity, genotoxicity, bond strength and microhardness of eight endodontic cements in one year evaluation

Abstract

In an attempt to find a material with suitable biological and physico-chemical properties, this study will determine in vitro genotoxicity and cytotoxicity by MTT assay and micronucleus formation, respectively, of endodontic sealers Apexit Plus, Real Seal, Endo Rez, Roeko Seal, AH Plus, Endomethasone, Polifil and a bioceramic sealer named iRoot SP. The cements are evaluated immediately after mixing, 24 hours after that and also after extended periods of hardening of the same (72 h, 1 week, 1, 3, 6 months and after a year) on human cell culture from the pulp (HPC1).The bond strength of cements to root canal walls will be evaluated by push-out test, in addition, microhardness' materials will be determined by a microhardness machine (both tests 15 days and 1 year after the manipulation). The cellular response to materials will be evaluated using the Kruskal-Wallis and Dunn's test (p = 0.05%), physical-mechanical tests will be compared using analysis of variance and Tukey's test (p = 0.05%).
