Cytogenetic evaluation of smoking damage in oral mucosa

Abstract

Recent studies have highlighted the role of tobacco in oral carcinogenesis, and that the exfoliative cytology is an effective technique for evaluation of these patients. The aim of the present study is to conduct a detailed analysis of chronic smoking effect by analysis of epithelial maturation, aneuploidy, and evaluation of epigenetic changes caused by methylation of the DNA repair genes validating by immunohistochemistry and molecular biology. Citological material collected from patients of the Outpatient Treatment Program Smoking Heart Institute, Hospital das Clinicas, Faculty of Medicine, University of São Paulo (INCOR-HCFMUSP). The sample will be studied in 5 groups as follow: Group 1: 40 initials smokers and smokers in tobacco treatment, Group 2: 40 nonsmokers in outpatient care in the Discipline of Stomatology FOSJC-UNESP, Group 3: group of 40 individuals treated with anti-smoking abstinent for 01 year. Groups 2 and 3 will be matched with group 1on the basis of age and sex. Group 4 will consist of 10 smokers, only male, that have not initiated the anti-smoking treatment and the Group 5 will consist of 10 nonsmokers equivalent sex and age group 4. Patients will be evaluated by history and the degree of dependence on nicotine and carbon monoxide level. Exfoliative cytology from the tongue and mouth floor will be performed. Smears will be evaluated for epithelial maturation, evaluation of aneuploidy, methylation profile analysis validated by quantitative RT-PCR in real time and protein analysis by immunohistochemical staining for hMLH1, hMSH2, ATM, CCNH. Statistical analysis will be conducted through descriptive analysis, using analysis of variance (ANOVA), Kruskal-Wallis and Mann-Whitney test, with significance level of 5%. (AU)
