Osteoarthritic chondrocytes and mesenchimal stem cells: relationship between inflammatory molecules and extracelullar matrix


Articular cartilage is a tissue of difficult healing. Osteochondral defects and osteoarthritis have a limited regenerative capacity. Usually, these diseases enhance tissue inflammatory cytokines and proteolytic molecules. It degrades extracellular matrix and causes a decreasing in articular cartilage function. Recently, stem cells are being studied in order to understand immunomodulatory features of these cells in tissue repair. The objective of our study is to determine effects of bone marrow stem cells on chondral cells, searching for anti-inflammatory and regenerative effects of stem cells. For this purpose, cultures of bone marrow stem cells and chondral cells from osteoartrhitic cartilage will be performed. Assessment of all cellular groups will be performed by microscopy for cell morphology. Inflammatory, catabolic, anabolic, and extracellular matrix molecules will be also assessed by immunohistochemestry and mRNA expression by real time PCR, ELISA of culture supernatant, and glycosaminoglycans will also be performed. Once pattern of these cells is achieved, coculture of bone marrow stem cells with osteoarthritic chondral cells will be performed. All assessments will be repeated after coculture to observe changes in cell and molecular pattern. We believe that bone marrow stem cells can change inflammatory cytokines and extracellular matrix pattern of chondral cells. Our results will be used as an in vitro basis for cell therapy for osteoarthritis. (AU)
