The process of cutaneous wound repair and re-epithelialization requires cellular and molecular events and can be divides into three main phases: inflammation, formation of granulation tissue with extracellular matrix deposition and remodeling. These phases occur simultaneously: they are superimposed. Diabetic patients cutaneous wound repair is impaired and there is not a single cause to the process impairment: failures or deregulation in cellular and molecular events are observed in the three phases of tissue repair. Recent studies have showed that insulin signaling, growth factors and cytokines have lower expression in wounds of diabetic rats. The search for new treatments has the purpose of accelerating wound healing and decrease risk of complications. Therefore, it is possible that isolated insulin or its association with others polypeptides growth promoters further different biochemical event, that collaborate to restore the tissue healing. The aim of this study will be investigate the effect of oral treatment, with atorvastatin and fenofribrate, and topical cream use, composed by insulin the wound healing and skin repair on the insulin signaling pathways, growth factors and cytokines of normal and diabetic rats. Male wistar will be used, eight-week-old rats. They will be weight and divided into four groups. The skin excision (6, 00 x 6,00 mm) or burn will be done in dorsal surface. In the 7th, 14th, 21st days after incision will be removed tissue samples for evaluation of protein expression and activation of signal transduction pathways by Immunohistochemistry methods, ELISA and immunoblotting. The results will be report as mean, standard deviation and the experiments numbers will be described. For statistical analysis, groups will be compared using two-way ANOVA with Bonferroni posttests. The results will be consider statistically significant when p< 0, 05. (AU)
