Evaluation of hematopoietic stem cell treatment in dogs with chronic renal failure

Abstract

There are currently few therapies available to prevent the progression of renal disease in humans or animals. To date, the only strategies that have been recommended are angiotensin-converting enzyme inhibitor use, blood pressure control, and a reduction in dietary protein. Chronic renal failure (CRF) in dogs is very similar to that seen in humans, especially in terms of the pathophysiological aspects, and CRF is being diagnosed with increasing frequency in dogs. Despite advances in the treatment of canine CRF, animals reaching the advanced stages of the disease are routinely euthanized. In a recent study involving an experimental rat model of CRF (5/6 nephrectomy), we showed that treatment with lineage-negative hematopoietic stem cells protects against CRF. In that study, lineage-negative hematopoietic stem cells were injected into the femoral vein on postnephrectomy day 15. On postnephrectomy day 60, treated rats presented reductions in proteinuria, glomerulosclerosis, anemia, renal infiltration of immune cells, and protein expression of monocyte chemoattractant protein -1. On postnephrectomy day 120, renal function (inulin clearance) was significantly better in treated rats than in untreated rats. The hypothesis of the present study was that administration of lineage-negative hematopoietic stem cells can slow the progression of CRF in dogs with stage 2 or 3 disease. The lineage-negative hematopoietic stem cells will be isolated from human umbilical cord blood. (AU)
