Subcutaneous tissue response of mice to triantibiotic paste indicated as intracanal medication for teeth with pulp necrosis and incomplete root formation

Abstract

Recently an alternative technique has been recommended for the treatment of teeth with incomplete root formation and pulp necrosis. A triantibiotic paste (TP) consisting of ciprofloxacin, metronidazole and minocycline is used for disinfecting root canal system followed by the induction of bleeding to fill the root canal and the coronal sealing. This technique allows root canal revascularization and root formation through the normal development of the thickness, length and morphology of the root. However there is no knowledge about the effect of TP on the connective tissue. This study will evaluate the gene expression of pro- and anti-inflammatory cytokines, hypoxia-induced factor (HIF),vascular endothelial growth factor (VEGF), metalloproteases (MMPs) and their inhibitors (TIMPs) induced by this paste. Forty mice will be distributed into four groups for each experimental period of 7 and 21 days. Each animal will receive 2 polyethylene tube implants containing the substances to be evaluated in the dorsal area: Group I - TP, Group II - calcium hydroxide paste (control), Group III - empty tube and Group IV - animals that will receive no tubes. After experimental periods of 7 and 21 days the tissue "capsule" present at the open end of the tube will be removed, followed by extraction of total RNA and preparation of complementary DNA. Detection and quantification of IL-1², TNF-±, IL-17, TGF-², VEGF, HIF, MMP-2 and -9 and TIMP-1 and -2 will be done through real-time PCR. It is our expectation to provide information about TP biological action and subsidies for the ideal intracanal medication selection to be used in permanent teeth with incomplete root formation and pulp necrosis.
