Development and validation of chromatographic and electrophoretic methods for subsequent application in studies of in vitro metabolism and biotransformation

Abstract

The quantification of analytes present in several biological matrices requires the development of methods that should be precise, accurate and reproducible under varying conditions of analysis. The high performance liquid chromatography and capillary electrophoresis techniques are widely used for these purposes. These techniques have several advantages such as: simplicity, speed of the analysis and, great versatility. Our research group has been working on the development and validation of chromatographic and electrophoretic methods to be further applied in studies of biotransformation and in vitro metabolism. The studies follow two distinct lines: (1) In vitro metabolism studies of potentially therapeutic compounds using rat liver microsomes and (2) use of fungi in the biotransformation of drugs, whose metabolism yields in chiral and pharmacologically active metabolites that are already marketed as drugs. In this context, it will be developed and validated bioanalytical methods for these purposes. This project aims to continue the work developed in the FAPESP regular project n. 2009/06132-0 which expires on September 30, 2011, but conducting studies with different substrates. It also seeks to implement and solidify a new line of research in the Chemistry Department of Faculdade de Filosofia, Ciências e Letras de Ribeirão Preto, developing studies of in vitro metabolism of compounds that have shown excellent pharmacological activities, using to this end, liver microsomes.
