Physical-chemical, mechanical, biocompatibility and bioactivity evaluation of different formulations of Portland cement and associations with micro and nanoparticles radiopacifying agents

Abstract

The Mineral Trioxide Aggregate (MTA) has Portland Cement (PC) in its basic composition. Additives and vehicles may be added to the CP to improve their consistency and applicability. The purpose of this study is to assess physical, mechanical and biological properties of White CP, and 4 different formulations and possible associations. White CP and CPs modified by additives will be evaluated using the following tests: setting time, flow, compressive strength, analysis of heavy metals, cellular viability (MTT), cell morphology assays, apoptosis / necrosis and comet test to assess possible genotoxic damage in cellular DNA. Two of the evaluated materials will be selected for a new phase of evaluations of the selected materials with different radiopacifying agents - zirconium oxide, niobium oxide, zinc oxide in micro and nanoparticles and bismuth oxide in microparticles. The CP and White MTA-Angelus will be used as a control. For the physical, chemical and mechanical tests, the methods will be: compressive strength, setting time, radiopacity, evaluation of surface, measurement of pH and calcium ion release and solubility. For biological analysis, the following tests will be made: MTT, cell morphology, necrosis / apoptosis, comet assay, cell cycle, evaluation of alkaline phosphatase, calcium deposits by Alizarin red dye and bioactivity of the materials plus the different radiopacifying agents. The data will be submitted to ANOVA and Tukey's test, with significance level of 5%. (AU)
