Abstract

Permeation studies in vitro and ex vivo performed in Franz diffusion cells are important during pre-clinical phase of a new developed medication because it is possible to preview how this drug will permeate through the barrier, i.e. mucosal or skin, allowing a pharmacokinetics and a biodisponibility improvement. The literature has shown that permeation studies across oral mucosa are performed using pig esophagus mucosa model. However, this tissue has little or no keratinization being the esophagus a limited model to evaluate the performance of formulations for dentistry use. There are other regions of the mouth with different levels of keratinization (palatal mucosa and gingival) which are also target sites to drugs such as local anesthetics, antimicrobial agents, corticoids, antivirus, etc. The objectives of the present study are: a) development and standardize different barriers to perform permeation studies (in Franz diffusion cells) of topical formulations for dentistry use; b) histological assessment of those barriers; c) to compare permeation of lidocaine across the different barriers in Franz diffusion cells; d) to evaluate freezing effect on the permeability and retention of those tissues. We hope to develop methods that could be useful for permeation studies for drugs used in dentistry, to improve their pharmacokinetic and pharmacodinamic performance. (AU)
