Abstract

Many studies have shown variations in cellular functions, anatomical structures and molecular level in aging. The non-enzymatic glycation of proteins (AGE) seems to be a common factor to the pathophysiology of a number of age-related disorders. Dermal equivalent is an efficient model for the study of cells and caused changes in the extracellular matrix in the aging process. The appearance of the products of AGEs occurs through reactions of non-enzymatic glycation of extracellular matrix in the dermis and has been touted as one of the factors responsible for the loss of elasticity and other properties of the dermis during aging. Drug penetration is a major constraint to compounds that are expected to reach deeper layers of skin. The 3D model of artificial skin has metabolic characteristics, permeability and activity similar to the original skin, increasing its role in research on drug permeability, toxicity, irritation and differentiation of keratinocytes. A series of natural compounds or synthetic inhibitors of AGEs have been proposed recently discovered or presented and may have therapeutic use in the treatment of changes caused by the formation and accumulation of AGEs in the skin. This study aimed to evaluate the efficacy and toxicity of the compound resveratrol, Carnosine, ALT-711, benfotiamine and pyridoxamine in glycated artificial skin model. (AU)
