Detraining is characterized by numerous physiological changes such as rapid weight gain, increased fat mass and insulin resistance, leading to a chronic low-grade inflammatory process. Studies have indicated that caloric restriction is an excellent nutritional strategy, reducing body fat, improving insulin sensitivity and reducing the risk of chronic inflammations. On the other hand, when associated with sedentary lifestyle, it accelerates muscle catabolism. In this respect, research has shown the beneficial effect of leucine supplementation in the maintenance of muscle mass, especially in situations of caloric restriction or nutritional recovery. Despite these facts, there is no evidence that the deleterious effects caused by detraining may be attenuated by caloric restriction associated with leucine supplementation. Thus, the aim of this study is to investigate the effects of caloric restriction and leucine supplementation on markers of inflammation and expression of proteins involved in the insulin signaling pathway and protein synthesis in untrained rats. For this study, 64 male adult Sprague-Dawley rats will be used. Initially, the animals will be distributed into two groups: control (CON) and Training [(TRAIN - animals will be trained on a treadmill]. Then, trained animals will be subdivided into five groups: Training (TREIN), Detraining (DT), Detraining + Caloric Restriction (DTRC), Detraining + Leucine (DT + LEU), Caloric Restriction + Detraining + Leucine (DTRC + LEU). We will analyze genes and proteins involved in protein synthesis and insulin signaling pathways (IRS-1, PKC, PKB, mTOR, p70S6K, IKK-beta; JNK; 4E-BP1), as well as markers of inflammation (IL-6, IL-10, MCP-1, TNF-±, 1L-1±, PAI-1, leptin; adponectin), glucose homeostasis and body composition. ANOVA will be used for comparison between groups, followed by Tukey test, considering a significance level of 5%. (AU)
