Influence of 5-fluorouracil in the normal periodontal tissues and the evolution of experimentally induced periodontitis

Abstract

The am this study to evaluate the histomorphometric and imunistoquimic the effects of 5-fluorouracil on normal periodontal tissues and on the development of experimentally induced periodontal disease (PD) and its influence on the mechanical periodontal treatment in rats. Will be used 150 animals were randomly divided into five groups of 30 animals which will be submitted to the following treatments: Group I - systemically healthy animals induced with PD, without local treatment. The animals of group no local or systemic treatment; Group II - systemically healthy animals induced with PD, animals treated locally with scaling and root planing (SRP). The animals of group no systemic treatment; Group III - animals treated systemically with 5-fluorouracil (5-FLU), without induction of PD and no local treatment; Group IV - animals with PD, the animals of this group will be treated systemically with 5-FLU and no local treatment; Group V - animals treated systemically with 5-FLU, induced PD and animals treated locally with SRP. The PD will induce for adaptation of a cotton ligature around the first molars. Animals systemically modified by the drug 5-fluorouracil (Fluorouracil, 50 mg / ml) will be receive the drug administration in two moments: immediately and 48 hours after removal of the ligature. The animals will be euthanized 10 animals in each group at 07,15 and 30 days after induction of treated PD. The specimens will be embedding in paraffin. Serial sections of 6 microns in thickness are obtained in the mesio-distal and stained with hematoxylin and eosin or Masson. Bone loss in the furcation region will be assessed by histomorphometric analysis and imunoistoquimic (RANKL, OPG e TRAP). Hemogram and level of alkaline phosphatase will be assessed. Data will be analyzed statistically (p <0.05) in specialized computer program (BioStat). (AU)
