Study of inflammatory modulators and treatment of mares resistant and susceptible to endometritis using platelet rich plasma


Breeding-induced endometritis (BIE) is the main cause of fertility reduction in mares. Recent reports have been shown the importance of inflammation immunomodulation after mate. The aim of this study is to verify the expression of inflammatory mediators in uterus before and after artificial insemination (AI) to find out the stimulus of semen on uterine inflammatory response, and, moreover, to propose a new treatment with the use of platelet rich plasma (PRP), which has been abundantly utilized because of its anti-inflammatory action. 15 mares resistant to BIE and 15 susceptible will be selected based on their reproductive index and ultrasonographic and cytological exams. The expression of endometrial mRNA of pro-inflammatory cytokines interleukins 1² (IL-1²), IL-6 and IL-8 and anti-inflammatory IL-10, Nitric Oxide (NO) dosage and the quantity of neutrophils on uterine secretion and exfoliative cytology will be analyzed in both group of animals; on the first cycle, there will not have drug intervention, and on the second, mares will be treated with 50mL of Platelet Rich Plasma administrated 4 hours after AI. The expression of such mediators will be analyzed in (1) under normal conditions and (2) 24 hours after AI with fresh semen. The expression of IL will be analyzed by real time-PCR, NO dosage by colorimetric reaction of Griess in spectrophotometer and the neutrophil in secretion will be quantified using Neubauer chamber. With this study, it is expected that treatment with PRP shows efficiency in immunomodulate the inflammatory response, as well as accelerate the resolution of inflammation that installs immediately and persists in certain mares after mate or AI.
