Effect of flouride on bone healing in the alveoli of mice with different bone densities: morphometric and zymography analysis

Abstract

Among the various proposed therapies for the treatment of osteoporosis, the administration of high doses of fluoride is an alternative that still has controversy in the literature. The fluoride is distinguished by influencing the activity of osteoblasts, which are responsible for mediating the synthesis of bone. However, the mechanism of action of fluoride on bone tissue cells (osteoblasts, osteocytes and osteoclasts) is not clear. Thus, this study aims to evaluate the alveolar bone repair in mice under the effect of fluoride administered in drinking water. In this work we will use four groups of mice of strains C57BL/6J (total 56) and C3H/HeJ (total 56) 45 days of life, which will receive water and deionized water with 50 ppm fluoride. Surgical procedures will be done by extracting the right upper incisor and the animals will be euthanized after 7, 14, 21 and 28 days. Immediately after euthanasia will be a cardiac puncture to obtain blood plasma for analysis of fluoride and obtaining the right number with dental socket. Will be made microscopic analysis (hematoxylin eosin staining), immunohistochemistry (ligand of receptor activator of nuclear factor kappa B: RANKL and osteoprotegerin, OPG) and protein for the repair of bone (matrix metalloproteinase 2 and 9: MMP-2 and 9 ) by Gelatin zymography technique. Statistical analysis will be made by the statistical test one-way ANOVA complemented by Tukey test, Kruskal-Wallis complemented by Dunn's test and will be at the "t" test, according to his statement (p <0.05).
