Bovine oocyte maturation: the influence of different siganling pathways


Although much advance has been achieved in research on in vitro production (IVP) of embryos, in particular in cattle due to its economical interest, embryo production is still beyond the desireable when compared with in vivo produced embryos interms of quantity and quality. IVP is constituted of different steps involving oocyte in vitro maturation (IVM), followed by in vitro fertilization (IVF) and then in vitro culture (IVC) of the embryos prpduced until they can be transferred to an appropriate recipient. Although all steps are important to the process, IVM is certainly crucial for final results in IVP. The present project unites three distinct blocks that aim to better understand a series of processes important for the development of oocyte quality in order for it to acquire in vitro its full potencial to generate good embryos after IFV and IVC. In Block I the aim is to establish a methodology for gene silencing by lipofection in cumulus cells as a tool for functional evaluation of genes expressed in cumulus cells during IVM and to investigate the importance of the BMPRII gene, expressed in cumulus cells during IVM, ,on oocyte competence. In Block II, it is proposed to investigate the contribution of different PDE isoforms on cAMP levels and meiosis resumption in bovine oocytes, and also to determine whether there are differences in signaling pathways in induced x spontaneous maturation and theis influence on oocyte competence. In Block III, the importance of melatonin on bovine oocyte in vitro maturation and embryo development, its possible role in cell protection (antioxidant/antipoptotic) and as an inducing fator for cell signaling pathways in the maturation of bovine cumulus-oocyte complexes.
