The cell culture system has been routinely used to evaluate cellular and MEC events of bone formation under controlled environment. In addition to osteoblast lineages, primary cultures of osteogenic cells derived from rat calvaria are also used. In a previous study carried out in our laboratory, dexamethasone-Dex (10-7 M) and TGF-beta 1 (5 ng/ml) were separately and simultaneously tested in osteogenic cells derived from rat calvaria. In all treatments the proteins OPN, BSP and FN were expressed. Only cells treated with Dex formed mineral nodules; however, in most cases, they were among few collagen fibrils and/or deposited on the substrate, suggesting that spontaneous nucleation of the mineral took place. When cells were treated with TGF-beta 1, changes in cell morphology and absence of ECM components were observed. Moreover, when treated with the two molecules simultaneously, cells were morphologically similar to that found in cells treated only with TGF-beta 1, but some mineralization foci were observed in collagen fibrils. The present project proposes to investigate the molecular effects of TGF-beta 1 and Dex, blocking receptors and studying some elements of their signaling pathways as well as their performance in the differentiation of osteogenic cells at different biomineralization stages by biochemically determine the alkaline phosphatase activity, histochemically detect mineral nodules, as well as by immunodetecting the expression of proteins such as collagen, OPN, BSP, OC, and ON by confocal microscopy. In addition, quantification by Western blot and RT-PCR will be also carried out. (AU)
