Abstract

The present project aims the characterization of Saccharomyces cerevisiae ORFs still identified with unknown function. Cells depleted for most of these ORFs do not show any clear phenotype, making the study of their respective function more problematic. However, our previous works in this subject were very profitable opening new interesting projects, such as those involved in coenzyme Q biosynthesis. These studies will also continue in the next two years. For instance, COQ8 over-expression will be tested in other CoQ mutants as well in cell cultures, looking for possible benefits of increasing the endogenous amount of CoQ as a consequence of extra Coq8p. Secondly, more studies involving the oxidative stress generated in these CoQ mutants are going to continue, focusing the effect in the stability of the mitochondrial DNA. And finally, we are going to use S. cerevisiae as a cellular model of neurodegenerative and neuromiopathies disorders. The SNCA human gene will be express in yeast cells in order to mimic the intracellular effects of parkinsonian neurons, and the respective consequences will be evaluate in the presence and absence of natural antioxidants.
