Evaluation stage of partition of the methanol extract of secondary metabolites isolated from Baccharis microdonta on pharmacological assays and enzyme activity of whole venom of Bothrops jararacusu and its PLA2

Abstract

The objective of this project is to evaluate the effect of phase partition of the methanol extract isolated from Baccharis microdonta and its metabolites on the pharmacological activities induced by venom of Bothrops jararacussu and its basic catalytically active sPLA2 (which will be isolated in the project). The tests to be evaluated are: edema, myotoxicity, platelet aggregation and thrombin like activity induced by whole venom. This assessment will be undertaken in two stages. At first, the different phases of partition and the metabolites are incubated with the whole venom, which after an hour, will be applied in animals. On Monday, a pre-treatment with the phases of partition and its metabolites will be performed by intraperitoneal injection (pre-set doses after LD50) one hour before injection of the venom. Beside the pharmacological evaluations also intend to investigate the effect of these compounds on enzymatic activity of phospholipase A2 and protease of the poison, using specific substrates BAPNA and BAADT for characterization of serine proteases, NGPANA to thrombin, and ADI-907-002 and Nobah for phospholipase A2. Additionally, this project will also verify the action of these compounds on the hemorrhage caused by whole venom metalloproteases and indirectly on the venom of Bothrops. (AU)
