Thrombogenic potential and gentamicin pharmacokinetics in plasma and synovial fluid of horses after administration in single doses, intravenous and intravenous regional

Abstract

The regional intravenous perfusion is a method that increases the concentration of antibiotic in infections that affect the distal portion of the members, or below the height of the radius, carpus and tarsus, contributing for illness elimination. This technique uses low doses of antibiotics compared with systemic therapy using the same drug, increasing the effectiveness while minimizing the risk of toxicity. The technique involves local delivery of antibiotics in distal limb tissues through the venous system, using superficial veins through a tourniquet placed proximal to the infected tissue, occluding the venous flow, than antibiotic is retained in the area infected. The objective is to evaluate the thrombogenic potential and gentamicin pharmacokinetics in plasma and synovial fluid of horses after administration in single doses, intravenous and intravenous regional. Blood samples from jugular vein and the carpal synovial fluid of all animals will be collected immediately before and 22 minutes, 45 minutes, 1 hour and 22 minutes, 2, 3, 4, 6, 8, 12, 24, 48, 72, 96 and 120 hours after drug administration. The concentration of gentamicin is measured by the method of Elisa and thrombogenic potential, by measuring plasma levels of Fibrin Degradation Products (PDF), D-Dimer, Antithrombin III, Doppler Ultrasound and thermography. Later the results will be compared statistically. (AU)
