Development of methodologies for photodynamic therapy applications on fungal infections

Abstract

The aim of this research project is to develop methodologies to study the photodinamic inactivation of Candida albicans in induced fungal infections in mice. Although several in vitro studies have demonstrated positive antifungal activities of photodynamic therapy, the application of this treatment in vivo requires adjustments of the conditions established in vitro. To reach the purpose of the present project, a mice model of vaginal Candida infection will be developed, in which photosensitizer concentration for its adequate distribution in tissue, irradiation parameters for effective antifungal effect, photodynamic inactivation of mutant C. albicans with overexpressed characteristics of drug resistance and the association of photodynamic therapy to drugs that inhibit those characteristics will be evaluated. In addition, the alterations in virulence of C. albicans induced by oxidative stress, after sublethal photodynamic therapy, will be investigated through in vitro tests and in vivo pathogenicity assay.
