Effect of lycopene supplementation on obesity in experimental model Zucker rats

Majority of researchers with obesity have been shown harmful in antioxidant defense system in obese However, little is known about the evaluation of important dietetic antioxidant lycopene in obesity and in diseases associate with obesity. The study aims to evaluate the oxidative stress level and the effect of supplementation with lycopene in obesity using rats Zucker obese. The study will be conducted with 16 rats Zucker obese (ZO) and 8 rats Lean with 8 weeks of age. The rats Zucker obese will be distribute in Zucker obese (ZO) and Zucker obese supplemented with lycopene (ZO+Lyco). The rats of group ZO + Lyco will receive the lycopene during 21 weeks (from 9 to 30 week of age). In the same period the rats of group L and ZO will receive corn oil by gavage with volume equivalent of group ZO+Lyco (0.5 ml/Kg weight/day). The systolic pression, the weight of animals and the Lee Index will be evaluated weekly. The serum of all the animals will be submit the laboratorial evaluation (glucose, insulin, triglycerides and total cholesterol), the dosages of lycopene, antioxidant capacity (TAP), activity of nicotinamide adenine dinucleotide phosphate oxidase (NADPH-oxidase), DNA lesions (Comet assay) in 9ª, 13ª, 20ª, 30ª week. Only in 30ª week, when the animals will be sacrifice, in the adipose tissue will be evaluate the activity of NADPH-oxidase by fluorometric method. The heart, the liver, right kidney of animals will be remove and storage to dosage of total antioxidant capacity (TAP). Also, the renal function will be evaluated in all the experimental groups.It is expected that the results may contribute with informations about the oxidative stress level and the effect of supplementation with lycopene in the clinical and laboratorial manifestations of obesity for the better understanding of its effects on the DNA, NADPH-oxidase and oxidative stress, aimed at reducing damage to patients obese.
