Evaluation of renal function in offspring of rats subjected to sleep restriction during pregnancy

Abstract

Clinical and experimental studies have shown that sleep restriction results in a variety of adverse effects, including hypertension, decreased glucose control, development of type II diabetes mellitus, obesity and damage at the immune system. Taking into account that changes in maternal environment can lead to several functional changes in the newborn, the purpose of this study is to study the impact of sleep restriction during pregnancy on blood pressure and on renal function and morphology of the offspring at adult age. Female rats, with three months of age will be allocated with a mate to mate. The pregnancy will be confirmed by the presence of sperm in vaginal smear. After this stage, the females will be randomly divided into three groups: control (C), RST (restriction of the sleep during the entire pregnancy) and RS (sleep restriction between the 14th and 20th days of pregnancy). The sleep restriction will be performed by the method of multi-platform, during 20 hours daily. After birth, it will be selected 6 male offspring per mother, designated as CO (offspring from C), ORST (offspring from RST), and ORS (offspring from RS). At 2 months of age, blood pressure (BP) of the offspring will be measured by plethysmography, and at 3 months old, it will be measured: renal plasma flow, glomerular filtration rate, renal excretion of sodium and potassium, urine osmolality, urinary protein excretion, expression of sodium transporters in renal tissue, glomerular area and number of glomeruli per mm3.
