Abstract

Fluoride is an element in the daily lives of almost all Brazilian population, being used in water supply (Brazil) and also as therapy for osteoporosis (common in Europe). In some cases, stimulation promotes bone formation by increasing the volume and density of bone in some patients with osteoporosis. In addition, several studies have shown an imbalance in bone remodeling (increased osteoclast activity) as a risk factor for osteoporotic fractures due to increased bone loss and deterioration of microarchitecture, where this constant cyclical process of bone remodeling imbalance is reflected in several markers biochemists. In this sense, the analysis of biochemical markers in the face of exogenous stimuli (consumption of fluoride) can assist in understanding how these elements are able to influence the process of mineralization. The monitoring of metabolic disorders (cellular and molecular level) can also provide clues about the strength of certain individuals to treatment and the therapeutic efficacy, thereby guiding more specific targets for treatment. However, there are still some doubts about the effectiveness of fluoride in reducing the risk of fractures. The precise mechanism by which fluoride influences the bone formation and mineralization is not fully understood, and in which species and concentrations have ill effects. To investigate which biochemical markers are susceptible to treatment with fluoride, osteoblasts will be used derived from C3H/HeJ and C57BL/6J, two species of mice with high and low bone density, respectively. Through an in vitro approach, which will be checked role of fluoride in the behavior of osteoblasts (rate of cell death and degree of differentiation of osteoblasts) and modulation of factors responsible for the degradation of the extracellular matrix.
