Microbiological, structural, mechanical and physical evaluation of resilient materials modified by the addition of antimicrobial agents for denture stomatitis treatment

Abstract

This study aims to evaluate microbiological, physical and mechanical properties of resilient materials after the addition of antimicrobial agents for denture stomatitis' treatment. The antimicrobial action of some agents (nystatin, miconazole, ketoconazole, itraconazole, and chlorhexidine) on the biofilm of Candida specimens (C. albicans, C. glabrata, and C. tropicalis) over resilient materials for temporary relining of prostheses (Softone and Trusoft) will be evaluated according to the determination of minimal inhibitory concentration (MIC) and sub-inhibitory concentration (MIC50). The fungal biofilm will be formed over specimens (with or without the drug addition) in aerobical conditions at 37ºC using YNB broth. The biofilm analyses will be made using two methods: tetrazolium salt reduction-XTT and confocal microscopy. MIC and MIC50 will be determined by the metabolic activity of viable cells according to XTT method after biofilm formation (48 h) and after 14 days. Confocal microscopy will be made on specimens presenting MIC and MIC50. After the determination of these concentrations, it will be analyzed the pattern of incorporation of those drugs in resilient specimens by using scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS) analyses. In addition, specimens with or without (control) drug addition will be submitted to Shore A hardness, superficial roughness, tensile strength, and peel bond strength to a denture base acrylic resin. These analyses will be made after the specimens have been immersed in distilled water at 37ºC for 24 h, 7, and 14 days. (AU)
