Immunohistochemical analysis of autogenous bone grafts repair under low-level lasertherapy: study in rabbits

The great benefits brought by oral and maxillofacial rehabilitations, especially in the field of dental rehabilitation using end osseous implants have been stimulating the development of techniques and materials, as well as improving the comprehension of tissue repair mechanisms directly involved in the rehabilitation process. Despite the well-established practice in ridge reconstruction using autogenous bone grafts from different embryologic origins, the molecular mechanisms that guide the processes of incorporation and resorption of the grafted bones are still not fully known. For better understanding of these events, molecular biology is a major tool. The preset study aims to analyze the expression of proteins related to bone repair process, as cyclooxygenase-2 (COX-2), nitric oxide, and transcription factor Cbfa-1, by means of immunohistochemistry, during the incorporation of onlay intramembranous (calvaria) and endochondral (iliac crest) bone grafts in the calvaria of New Zealand rabbits, in the periods of 7, 14, 30 and 60 days, under continuous administration of specific COX-2 anti-inflammatory drug. Another experimental group in the same conditions will be submitted to low-level laser therapy immediately after the surgical procedure, continuing every 48 hour for 15 days, or until the day of sacrifice, in order to verify the expression of the same proteins under this specific condition. The results will be associated to the process of bone grafts repair focusing on new bone formation in the interface area, and maintenance of the grafts volume, by means of histomorphometry and direct measurement, respectively.
