Abstract

This project focuses the technological development of cationic liposomes carrying LACK DNA and shielded by hyaluronic acid as intranasal gene vaccine against Leishmaniasis. The in vivo and intranasal LACK DNA efficiency was demonstrated by the Instituto de Biofísica Carlos Chagas Filho-RJ. The hyaluronic acid is a natural polisacaride with mucoadesive properties. It had been used in intranasal vaccines and it also increases the DNA delivery into cells when incorporated in cationic liposomes. The liposomes will be composed by egg phosphatidylcholine, L-alpha-dioleoyl phosphatidylethanolamine and 1,2-dioleoyloxy-3-trimethylammonium propane. The hyaluronic acid that will be used for liposomes shielding will have low molecular weight. The physico-chemical characterization will be zeta potential, morphology, mean diameter, polydispersity and size distribution. The LACK DNA incorporation will be evaluated using fluorescence probe accessibility, molar charge ratio for complete DNA incorporation into the liposome, plasmid integrity after processing, using agarose gel electrophoresis technique. The best formulation will be produced and send for in vivo evaluation at the Instituto de Biofísica Carlos Chagas - RJ. Our previous experience with cationic liposomes focused the development of a DNA vaccine against tuberculosis (one patent, one addition certificate and 4 publications) and scale up studies for liposome production and its complexation with DNA (FAPESP PIPE II). In this context, this project will contribute to the development of new nanostructures useful for intranasal gene vaccination against Leishmaniasis. (AU)
