Effects of photodynamic therapy and essential oils of the Cymbopogon citratus (DC.) Stapf, Casearia sylvestris SW and Curcuma zedoaria Roscoe, in vitro, in biofilms of Candida albicans, Staphylococcus aureus and Streptococcus mutans

Abstract

The aim of this study is to evaluate the effects of photodynamic therapy (TFP) and antimicrobial activity of essential oils from Cymbopogon citratus (DC.) Stapf, Curcuma zedoaria Roscoe and Casearia sylvestris SW in the development of biofilm, produced by C. albicans, S. aureus and S. mutans and associations. In the assessment of essential oils effects and photodynamic therapy, a biofilm will be produced with standard microorganisms of C. albicans (ATCC 18804), S. aureus (ATCC 6538) and S. mutans (ATCC 35688) in acrylic disks for 5 days in sterile brain heart infusion both (BHI) containing 5 % sucrose. After this time it will be performed the photodynamic therapy with AsGaAl laser and photosensitivity methylene blue and the essential oils will be tested by a period of 5 minutes. The biofilm will be dispersed by ultrasound and serial dilutions will be carried out and aliquots plated in selective agar to the numbers of colony-forming units by milliliters (CFU/mL). Also, the phytoterapy and TFP effects will be evaluated in microrganisms of planktonic form. For the minimal inhibition concentration (MIC) of the essential oils it will be utilized 10 C. albicans, 10 S. aureus and 10 5. mutans clinical samples. The minimal inhibition concentration (MIC) will be made by serial dilutions. The result will be converted in logarithms of base 10 (logio) and analyzed statistically by the t Student (p³0,05) test. To illustrate the adherence of microorganisms C. albicans, S. aureus and S. mutans and associations a Scanning Electron Microscopy images (SEM) will be performed. (AU)
