Evaluation of Artin M on wound healing in palatal mucosa: an in vitro and in vivo study

Abstract

Artin M, a lectin from Artocarpus integrifolia seeds, has been shown to accelerate the process of wound healing on burn injuries and corneal epithelial lesions in rabbits. The aim of this study is to evaluate the effects of Artin M on wound healing in palatal mucosa in vivo and in vitro. In vivo: Seventy-two rats will be divided into three groups according to one of the treatment assigned: C - Control (non-treated), A - Artin M, and V - Vehicle. A 4mm full thickness wound will be surgically created on palatal mucosa of each animal. Eight animals per group will be sacrificed in each time point of the 3, 5, and 7 days post surgery. Maxillary biopsy will be taken and divided to perform: 1- histological analysis: to evaluate area of reepithelization, morphology of granulation tissue, the number of inflammatory cells, maturation of the collagen fibers and neovascularization. 2-Immunohistochemistry analysis: immunostaing for macrophages, bFGF, VEGF, TGF²) and, PCNA. 3- Protein expression: of MPO activity VEGF and TGF². In vitro: Rat gingival fibroblasts (primary culture) and macrophages from rats will be cultured in the presence of different concentration of Artin M (0.01, 0.1 or 1 ¼g/ml) for different periods (4, 8, 12 e 24 h) at 37ºC, 5% CO2 to evaluate gene and protein expressions of growth factors (VEGF, TGF²) and proinflammatory cytokines (IL-1 and TNF±).
