Expression and putative roles played by Sdf2 during gestation

Abstract

Using a signal sequence trap method (SST) Hamada et al. (1996) have isolated the cDNA encoding of a novel secretory protein from a stroma cell line ST2 that can support differentiation of hematopoietic cells (Ogawa et al., 1988). They determined the complete nt sequences of this mouse and human protein nominating it stroma derived factor 2, SDF-2. The amino acid (aa) sequence deduced from the nt sequence of SDF-2 cDNA predicts that SDF-2 is likely a secretory protein but not a membrane protein. Genomic Southern blot analysis indicated that the SDF-2 gene is relatively conserved among mammals. Hybridized bands were detected in rabbit, mouse and human DNA, but not in chicken and X. laevis DNA. Through macroarray cDNA approaches (GE Array O Series, MM-15 mouse inflammatory cytokines & receptors gene array, 96 genes), we first analyzed the transcription of chemokines and its receptors in freshly isolated mouse ectoplacental cone cells, a pure trophoblast cell population. Among 96 chemokines and receptors, Sdf-2 was found highly transcribed (Hoshida, 2007). Through macroarray cDNA approaches (GE Array O Series, MM-15 mouse inflammatory cytokines & receptors gene array, 96 genes), we first analyzed the transcription of chemokines and its receptors in freshly isolated mouse ectoplacental cone cells, a pure trophoblast cell population. Among 96 chemokines and receptors, Sdf-2 was found highly transcribed (Hoshida, 2007). Thus, based on the following facts: i) chemokines may play important roles in hematopoiesis, cell proliferation, cell differentiation and migratory processes; ii) trophoblast cells perform endovascular invasion, acquire endothelial cell-like phenotype and position themselves in direct contact with maternal blood flow and, thus, are possible candidate to attract immune competent cells such as NK cells or dendritic cells from peripheral blood to the decidua; iii) endovascular trophoblast may contribute with endothelial cell migration and proliferation for angiogenic purposes at the maternal compartment of the placenta; iv) trophoblast cells establishes close contact with decidual cells to form the decidual-chorionic unit and therefore may participate of endometrial cell viability through the activation of specific signaling pathways.Therefore, it is interesting to speculate whether chemokines produced at the maternal-fetal interface may interact with chemokine receptors expressed by surrounding cell types so as to regulate the growth, differentiation, and functioning of the placenta in paracrine or autocrine manners. The developmental pattern of Sdf-2 distribution and expression may provide some useful insights into the cellular mechanisms of embryo implantation. In the present study, we would like to analyze the secretory protein Sdf-2 gene and protein expression and, functional roles during peri-implantation period using RT-PCR, qPCR, immunohistochemistry, western blot, oligodeoxynucleotides and assays of invasion, proliferarion and cell death.
